Abstract

OBJECTIVE

To analyze the effect of an NY-ESO-1 expressing HCC cell line by NY-ESO-1b specific CD8(+)T cells in vitro induced by HLA-A2 Restricted NY-ESO-1b peptide and to further analyze the possibility of NYjESOj1 as a vaccine in HCC treatment.

METHODS

Peripheral blood mononuclear cells (PBMCs) were obtained from an HLA-A2(+) HCC patient and NY-ESO-1 specific CD8(+)T cells were in vitro induced by HLA-A2 Restricted NY-ESO-1b peptide. Next we estimated the effect that NY-ESO-1 specific CD8(+)T cells recognized and killed NY-ESO-1 expressing HepG2 cells by enzyme-linked immunospot(ELISPOT).

RESULTS

These in vitro induced NY-ESO-1b specific CD8(+)T cells could recognize HepG2 cells stably transfected with NY-ESO-1 in both IFN-gamma and Granzyme B ELISPOT assays. And the data also showed that NY-ESO-1 specific CD8(+)T cells could efficiently kill NY-ESO-1 expressing HepG2 cells.

CONCLUSION

NY-ESO-1 protein can be processed by HCC cells, and that HLA-A2 restricted NY-ESO-1b peptide (157-165) can also be presented on the surfaces of cells and recognized by NY-ESO-1b specific CD8(+)T cells in vitro induced.

目的

分析由 HLA - A2 限制性 NY - ESO - 1b 肽体外诱导的 NY - ESO - 1b 特异性 CD8(+)T 细胞对表达 NY - ESO - 1 的肝癌细胞系的作用,并进一步分析 NY - ESO - 1 作为肝癌治疗疫苗的可能性。

方法

从一名 HLA - A2(+)肝癌患者获取外周血单个核细胞(PBMC),并用 HLA - A2 限制性 NY - ESO - 1b 肽体外诱导 NY - ESO - 1 特异性 CD8(+)T 细胞。接下来,我们通过酶联免疫斑点法(ELISPOT)评估 NY - ESO - 1 特异性 CD8(+)T 细胞识别并杀伤表达 NY - ESO - 1 的 HepG2 细胞的效果。

结果

结论

NY - ESO - 1 蛋白可被肝癌细胞加工处理,且 HLA - A2 限制性 NY - ESO - 1b 肽(157 - 165)也能呈递在细胞表面,并被体外诱导的 NY - ESO - 1b 特异性 CD8(+)T 细胞识别。
