Abstract

OBJECTIVE

To observe the effects of Kangfengshi Granules (KFSG) on expressions of the mRNAs of osteoprotegerin (OPG), receptor activator of nuclear factor-kappaB ligand (RANKL) and macrophage colony stimulating factor (M-CSF) in bone tissues of rats with collagen-induced arthritis.

METHODS

Forty SD rats were randomly divided into four groups: normal control group, untreated group, cyclosporine A (CsA)-treated group and KFSG-treated group. Except the rats in the normal control group, all the other rats received subcutaneous injection of collagen II to establish collagen-induced arthritis (CIA) models. Then the rats in each group were fed normal saline or corresponding drugs for four weeks. Total RNA was extracted from carpal and digital bones. The expressions of OPG, RANKL and M-CSF mRNAs were examined by real-time PCR.

RESULTS

The total incidence of arthritis induced by collagen II in the rats was approximately 90%. The expression levels of RANKL and M-CSF mRNAs and the RANKL mRNA/OPG mRNA ratio in the untreated group, KFSG-treated group and CsA-treated group were all significantly higher than those in the normal control group, while the expression levels of OPG mRNA in those three groups were significantly lower than that in the normal control group. The expression level of OPG mRNA in the KFSG-treated group was obviously higher while the expression level of M-CSF mRNA and the RANKL mRNA/OPG mRNA ratio in the same group were both lower as compared with those in the untreated group.

CONCLUSION

The molecular mechanism of effects of KFSG on bone erosion and destruction induced by rheumatoid arthritis is closely correlated with up-regulating the expression of OPG mRNA, down-regulating the expression of M-CSF mRNA and RANKL mRNA/OPG mRNA ratio.

目的

观察抗风湿颗粒(KFSG)对胶原诱导性关节炎大鼠骨组织中骨保护素(OPG)、核因子κB受体活化因子配体(RANKL)及巨噬细胞集落刺激因子(M-CSF)mRNA表达的影响。

方法

40只SD大鼠随机分为四组:正常对照组、未治疗组、环孢素A(CsA)治疗组和KFSG治疗组。除正常对照组大鼠外,其余大鼠均皮下注射Ⅱ型胶原以建立胶原诱导性关节炎(CIA)模型。然后每组大鼠给予生理盐水或相应药物,连续喂养四周。从腕骨和指骨中提取总RNA。采用实时荧光定量PCR检测OPG、RANKL和M-CSF mRNA的表达。

结果

Ⅱ型胶原诱导的大鼠关节炎总发病率约为90%。未治疗组、KFSG治疗组和CsA治疗组中RANKL和M-CSF mRNA的表达水平以及RANKL mRNA/OPG mRNA比值均显著高于正常对照组,而这三组中OPG mRNA的表达水平显著低于正常对照组。与未治疗组相比,KFSG治疗组中OPG mRNA的表达水平明显升高,而同一组中M-CSF mRNA的表达水平和RANKL mRNA/OPG mRNA比值均降低。

结论

KFSG对类风湿关节炎所致骨侵蚀和破坏作用的分子机制与上调OPG mRNA表达、下调M-CSF mRNA表达及RANKL mRNA/OPG mRNA比值密切相关。
