To explore the effects of ligustrazine on cell proliferation in hippocampal dentate gyrus subgranular zone (SGZ) after focal cerebral ischemia in adult rats.
Middle cerebral artery occlusion (MCAO) model was established in adult rats by placement of an intraluminal filament at the origin of the MCA. Ligustrazine was administered intraperitoneally at a daily dose of 80 mg/kg starting at 2 h after MCAO, and BrdU (50 mg/kg daily) was also injected intraperitoneally starting at 4 h after MCAO. BrdU-positive cells in the SGZ were counted 7, 14 and 24 days after MCAO, respectively.
Compared with sham operation group, ischemic ipsilateral BrdU-positive cells in the ischemic model group increased 7 days after MCAO, reaching the peak on day 14, and decreased on day 21 (P<0.01). The number of ischemic ipsilateral BrdU-positive cells in ligustrazine group was significantly greater than that in the ischemic model group on days 7, 14 and 21 (P<0.01), and maintained the high level on day 21.
Ligustrazine possesses long lasting effect of promoting cell proliferation in the SGZ after focal cerebral ischemia in adult rats.
探讨川芎嗪对成年大鼠局灶性脑缺血后海马齿状回颗粒下区(SGZ)细胞增殖的影响。
通过在大脑中动脉起始部置入腔内细丝建立成年大鼠大脑中动脉闭塞(MCAO)模型。川芎嗪于MCAO后2小时开始腹腔注射,每日剂量80mg/kg,5-溴脱氧尿嘧啶核苷(BrdU,每日50mg/kg)也于MCAO后4小时开始腹腔注射。分别在MCAO后7天、14天和24天对SGZ中的BrdU阳性细胞进行计数。
与假手术组相比,缺血模型组缺血同侧BrdU阳性细胞在MCAO后7天增加,在第14天达到峰值,在第21天减少(P<0.01)。川芎嗪组缺血同侧BrdU阳性细胞数量在第7天、14天和21天显著多于缺血模型组(P<0.01),并在第21天维持在高水平。
川芎嗪对成年大鼠局灶性脑缺血后SGZ细胞增殖具有持久的促进作用。
