To study whether gastrin/cholecystokinin-B (CCK-B) receptor autocrine loop exists in human gastric cancer cells and the effects of gastrin/CCK-B receptor autocrine loop on the growth and apoptosis of human gastric cancer cells.
Human gastric cancer cells of the line SGC-7901 were cultured. The expression of gastrin and CCK-B was detected by RT-PCR, immunocytochemistry and radioimmunoassay. Antibody against gastrin was added so as to block the autocrine loop to observe the growth rate, cell cycle and apoptosis by 3-(4.5-dimethylthiazol-z-yl) -2, 5-diphenyl tertrazolium blue (MMT) colorimetric assay and flow cytometry (FCM).
The SGC-7901 cells co-expressed gastrin and CCK-B receptor mRNAs, confirmed by sequencing, thus forming gastrin/the CCK-B receptor autocrine loop. After the blocking of the autocrine loop by antibody against gastrin, the cell growth rate and the number of cells residing in the S-phase of the cell cycle decreased and the apoptotic rate increased in an antibody concentration-dependent manner.
Human gastric cancer SGC-7901 cells possess the gastrin/the CCK-B receptor autocrine loop. Blocking the autocrine loop inhibits the cell proliferation and promotes the cell apoptosis.
研究胃泌素/胆囊收缩素-B(CCK-B)受体自分泌环路是否存在于人类胃癌细胞中,以及胃泌素/CCK-B受体自分泌环路对人类胃癌细胞生长和凋亡的影响。
培养人胃癌SGC-7901细胞系。采用逆转录聚合酶链反应(RT-PCR)、免疫细胞化学和放射免疫分析法检测胃泌素和CCK-B的表达。加入抗胃泌素抗体以阻断自分泌环路,通过噻唑蓝(MTT)比色法和流式细胞术(FCM)观察细胞生长速率、细胞周期和凋亡情况。
SGC-7901细胞共表达胃泌素和CCK-B受体mRNA,经测序证实,从而形成胃泌素/CCK-B受体自分泌环路。用抗胃泌素抗体阻断自分泌环路后,细胞生长速率和处于细胞周期S期的细胞数量减少,凋亡率以抗体浓度依赖性方式增加。
人胃癌SGC-7901细胞具有胃泌素/CCK-B受体自分泌环路。阻断自分泌环路可抑制细胞增殖并促进细胞凋亡。
