Abstract

Treatment of cytosol from the rat ventral prostate with cold acetone (-20 degrees C) evoked a 8 approximately 10-fold increase in the binding capacity with 5alpha-dihydrotestosterone (DHT). Starting from the extract of acetone-dried prostate cytosol, some 400 approximately 600-fold purification of the DHT-binding protein complex was acieved by (NH4)2504 fractionation, DEAE-cellulose chromatography and gel- filtration with Sephadex G-200. The purified 3H-DHT-binding protein complex was incorporated into the nuclei from the ventral prostate in a temperature dependent manner. 用冷丙酮(-20℃)处理大鼠腹侧前列腺的胞质溶胶,可使与5α-二氢睾酮(DHT)的结合能力提高约8至10倍。从丙酮干燥的前列腺胞质溶胶提取物开始,通过硫酸铵分级分离、DEAE-纤维素色谱法和Sephadex G-200凝胶过滤,实现了DHT结合蛋白复合物约400至600倍的纯化。纯化的3H-DHT结合蛋白复合物以温度依赖的方式掺入腹侧前列腺的细胞核中。在肝脏和肾脏的细胞核中也观察到类似的掺入情况……
