BACKGROUND & AIMS: Glutamine synthetase (GS), a key enzyme in the glutamine synthesis, is thus crucial in glutamine homeostasis. GS is known to be up-regulated by fasting and inhibited by glutamine supplementation. The aim of this study was to determine whether the presence of glucose in glutamine supplementation with refeeding differently affects up-regulation of muscle GS by fasting in vivo in adult female rats than glutamine alone.
Muscle GS activities were assessed in 5-day-fasted female Wistar adult rats refed and supplemented with glutamine or glycine in the presence or not of glucose.
After 5-day-fasting, the up-regulated GS activity was decreased whatever the type of amino acid supplementation (glutamine or glycine), whereas it was more decreased by supplementation with a mixture glutamine/glucose. In glycine/glucose supplemented rats, no effect of glucose supplementation was observed on GS activity.
These results demonstrated that intramuscular glutamine was spared when glucose was added to glutamine supplementation in adult rats. Consequently, the role of glucose consisted in slowing down the glutamine synthesis. By contrast, glucose has no role when it was associated with glycine whose degradation does not produce energy.
谷氨酰胺合成酶(GS)是谷氨酰胺合成中的关键酶,因此在谷氨酰胺稳态中至关重要。已知GS会因禁食而上调,并受谷氨酰胺补充剂的抑制。本研究的目的是确定在成年雌性大鼠体内,在再喂养时谷氨酰胺补充剂中添加葡萄糖是否比单独使用谷氨酰胺对禁食引起的肌肉GS上调有不同影响。
对禁食5天的成年雌性Wistar大鼠进行再喂养,并在有或无葡萄糖的情况下补充谷氨酰胺或甘氨酸,评估其肌肉GS活性。
禁食5天后,无论补充何种氨基酸(谷氨酰胺或甘氨酸),上调的GS活性均降低,而补充谷氨酰胺/葡萄糖混合物时降低得更多。在补充甘氨酸/葡萄糖的大鼠中,未观察到补充葡萄糖对GS活性有影响。
这些结果表明,在成年大鼠中,当谷氨酰胺补充剂中添加葡萄糖时,肌肉中的谷氨酰胺得以保留。因此,葡萄糖的作用是减缓谷氨酰胺的合成。相比之下,当葡萄糖与不产生能量的甘氨酸结合时则没有作用。
