To explore the effects of bears' bile on tumor cell p53 protein expression with different gene properties.
The effects of bears' bile on the expression of p53 protein in 6 cancer cell strains were determined by Western blot and reverse transcription-polymerase chain reaction (RT-PCR) analysis. Results Western blot analysis showed that the expression of p53 protein in HaCaT, KUMA3, KUMA4 and KUMA6 cell strains with gene mutation were increased, but no change was found in HCT116 and KUMA5 cell strains without gene mutation. 探讨熊胆对具有不同基因特性的肿瘤细胞p53蛋白表达的影响。
采用蛋白质免疫印迹法(Western blot)和逆转录-聚合酶链反应(RT-PCR)分析,检测熊胆对6种癌细胞株p53蛋白表达的影响。结果蛋白质免疫印迹法分析显示,HaCaT、KUMA3、KUMA4和KUMA6基因突变细胞株中p53蛋白表达增加,而HCT116和KUMA5无基因突变细胞株中p53蛋白表达无变化。采用RT-PCR分析系统分析p53 mRNA,所有细胞株中p53 mRNA均无定量变化。
熊胆对癌细胞株p53蛋白表达的影响可能因p53基因特性而异,即熊胆仅影响突变型p53蛋白。
