To investigate the intervenient effect of compound traditional Chinese medicine on dendritic cells (DCs) in peripheral blood in vitro.
DCs in peripheral blood were cultured for five days with medium which had GM-CSF and IL-4. The compound traditional Chinese medicine were added into the medium and the cells were collected on the 5th day. Surface markers of CD83 and CD86 in DCs were analyzed by flow cytometry. The capability of DC to stimulate the proliferation of T lymphocytes was evaluated by allogeneic mixed lymphocyte reaction. The content of IL-12 in cell culture supernatant was detected by ELISA.
The expression of CD83 and CD86 was increased significantly(P<0.001) after the addition of the compound traditional Chinese medicine. The capability of DC to stimulate the proliferation of T lymphocytes was increased evidently (P<0.05). However the production of IL-12 was decreased obviously(P<0.001).
The compound traditional Chinese medicine has immunoregulation of DC, It can enhance the presenting capability of antigen in DCs and inhibit the production of IL-12.
研究复方中药对体外培养的外周血树突状细胞(DCs)的干预作用。
用含粒细胞-巨噬细胞集落刺激因子(GM-CSF)和白细胞介素-4(IL-4)的培养基培养外周血DCs 5天。将复方中药加入培养基中,并于第5天收集细胞。采用流式细胞术分析DCs表面标志物CD83和CD86。通过异体混合淋巴细胞反应评估DC刺激T淋巴细胞增殖的能力。采用酶联免疫吸附测定法(ELISA)检测细胞培养上清液中白细胞介素-12(IL-12)的含量。
加入复方中药后,CD83和CD86的表达显著增加(P<0.001)。DC刺激T淋巴细胞增殖的能力明显增强(P<0.05)。然而,IL-12的产生明显减少(P<0.001)。
复方中药对DC具有免疫调节作用,可增强DC的抗原呈递能力并抑制IL-12的产生。
