To explore effects of Yishen Decoction on the expression of MMP-9 and TIMP-1 in kidney of mice with IgA nephropathy (IgAN).
The IgAN model was established by oral administration of bovine serum albumin (BSA) together with the injection of Staphylococcus enterotoxin B (SEB) through caudal vein. The mRNA and protein expression of MMP-9 and TIMP-1 were measured by fluorescent quantitative RT-PCR and immunohistochemistry respectively.
No significant differences of mRNA and protein expression of MMP-9 and TIMP-1 were found between the low and high concentration Yishen Decoction treated group. But the expression of TIMP-1 in the Yishen Decoction treated group was lower than that in the IgAN group, while that of MMP-9 was higher than that in the control group.
The TCM Yishen Decoction can inhibit the abnormal expression of TIMP-1, and strengthen the expression of MMP-9 in kidney of mouse with IgA nephropathy.
探讨益肾汤对免疫球蛋白A肾病(IgAN)小鼠肾脏基质金属蛋白酶-9(MMP-9)和基质金属蛋白酶组织抑制因子-1(TIMP-1)表达的影响。
通过口服牛血清白蛋白(BSA)并经尾静脉注射B型肠毒素(SEB)建立IgAN模型。分别采用荧光定量逆转录聚合酶链反应(RT-PCR)和免疫组化法检测MMP-9和TIMP-1的信使核糖核酸(mRNA)及蛋白表达。
低、高浓度益肾汤治疗组MMP-9和TIMP-1的mRNA及蛋白表达差异无统计学意义。但益肾汤治疗组TIMP-1的表达低于IgAN组,而MMP-9的表达高于对照组。
中药益肾汤可抑制IgAN小鼠肾脏TIMP-1的异常表达,并增强MMP-9的表达。
