To investigate the role of Smac in cisplatin-induced apoptosis of non-small lung cancer cells in vitro.
Non-small cell lung cancer A549 cells were incubated in the presence of cisplatin at different concentrations, and the cell proliferation status was observed using MTT assay. Flow cytometry was used for evaluation of the apoptosis of the incubated cells, and the expressions of Smac mRNA and protein were detected by RT-PCR and Western blotting, respectively.
Cisplatin inhibited the proliferation and induced apoptosis of A549 cells both in a concentration-dependent manner. Cisplatin also increased the expression of Smac at both the mRNA and protein levels, which was also concentration-dependent.
Increased Smac expression may play a critical role in cisplatin-induced apoptosis of the non-small cell lung cancer cells in vitro.
探讨Smac在顺铂诱导的非小细胞肺癌细胞体外凋亡中的作用。
将非小细胞肺癌A549细胞在不同浓度顺铂存在下孵育,采用MTT法观察细胞增殖状态。流式细胞术用于评估孵育细胞的凋亡情况,分别通过RT-PCR和蛋白质印迹法检测Smac mRNA和蛋白的表达。
