We performed a prospective randomized and controlled animal study to investigate the effects of infliximab on experimental otitis media in rats.
Seventy-two Sprague-Dawley rats were randomly allocated to 3 study groups and 1 control group. Infliximab was injected intravenously. Histopathologic changes were determined by hematoxylin-eosin staining. Fluorescence microscopy was performed to examine the leakage of the exudates. Vascular permeability was measured by the Evans blue dye technique.
In comparison with the control group, we found significant differences in the extent of middle ear mucosa without active inflammation and the presence of reparable lesions in all study groups treated with infliximab. A significant reduction of extravasated Evans blue dye in all study group animals was found as compared with the control group animals.
This study suggests that the monoclonal tumor necrosis factor alpha antibody, infliximab, can reduce inflammatory activity in experimental otitis media in rats.
我们进行了一项前瞻性随机对照动物研究,以调查英夫利昔单抗对大鼠实验性中耳炎的影响。
将72只斯普拉格-道利大鼠随机分为3个研究组和1个对照组。通过静脉注射英夫利昔单抗。采用苏木精-伊红染色确定组织病理学变化。进行荧光显微镜检查以检测渗出物的渗漏情况。采用伊文思蓝染料技术测量血管通透性。
与对照组相比,我们发现所有接受英夫利昔单抗治疗的研究组中耳黏膜无活动性炎症的程度及可修复病变的存在情况存在显著差异。与对照组动物相比,所有研究组动物渗出的伊文思蓝染料均显著减少。
本研究表明,单克隆肿瘤坏死因子α抗体英夫利昔单抗可降低大鼠实验性中耳炎的炎症活性。
