More than 150 years after multiple sclerosis (MS) was first described, its cause remains unknown. In defining the aetiology of MS the question of the relative importance of genetic and environmental factors in the development of the disease is crucial. Familial clustering of cases strongly supports the hypothesis that genetic factors are fundamental in determining disease susceptibility.
