To test the effect of RNA interference of hnRNP Bx gene in human lung cell line A549.
The RNAs of cell line A549 were interfered by the RNA interference technique. The hnRNP B1 mRNAs were detected by fluorescence quantity RT-PCR. The proteins of hnRNP B1 were detected by Western blot technique. The growth rate of the cells was analysed by MTT.
The growth of the cells was suppressed significantly. The mRNA and protein expressions of hnRNP B1 were inhibited.
The expression of hnRNP B1 can be inhibited by RNA interference. RNA interference could become a possible clinical therapy for lung cancers.
检测RNA干扰人hnRNP Bx基因对人肺癌细胞系A549的影响。
采用RNA干扰技术干扰A549细胞系的RNA。通过荧光定量RT-PCR检测hnRNP B1 mRNA。采用蛋白质免疫印迹技术检测hnRNP B1蛋白。用MTT法分析细胞的生长速率。
细胞生长受到显著抑制。hnRNP B1的mRNA和蛋白表达均受到抑制。
RNA干扰可抑制hnRNP B1的表达。RNA干扰可能成为肺癌的一种临床治疗方法。
