To investigate the role of recombinant human growth hormone (rhGH) in the growth of Bel-7402 human hepatic carcinoma cell line (Bel-7402 line) in vitro and its effects on GHR expression.
Tumor cell count, MT assay and colony forming test were performed to determine the responses of Bel-7402 to different concentrations of rhGH (0, 1, 10, 100, 1000, 10 000 ng/ml). Metabolism of DNA in tumor cells was analyzed with the method of mixture of 3H-TdR. Radioreceptor assay was used to detect the GHR expression of the hepatic carcinoma cell lines and its relation to different rhGH concentrations.
rhGH accelerated the proliferation of the Bel-7402 line when the concentration of rhGH was over 100 ng/ml (P < 0.05). Other rhGH concentrations had also positive effects, but with reduced effect as compared with that of 100 ng/ml. After 24 h of rhGH addition of concentration of 10 ng/ml and 100 ng/ml, GHR site number was significantly higher than that in control group, while the 10,000 ng/ml group showed a significantly lower GHR site number.
Different concentrations of rhGH might result in variable effects on the growth of Bel-7402 hepatic carcinoma cell line. Certain concentrations of rhGH might stimulate the growth of the cell line. rhGH can regulate the expression of GHR in the cell line.
探讨重组人生长激素(rhGH)对人肝癌细胞系Bel-7402(Bel-7402细胞系)体外生长的作用及其对生长激素受体(GHR)表达的影响。
采用肿瘤细胞计数、MTT法和集落形成试验,检测Bel-7402细胞对不同浓度rhGH(0、1、10、100、1000、10000 ng/ml)的反应。用3H-TdR掺入法分析肿瘤细胞DNA代谢。采用放射受体分析法检测肝癌细胞系的GHR表达及其与不同rhGH浓度的关系。
当rhGH浓度超过100 ng/ml时,可促进Bel-7402细胞系的增殖(P<0.05)。其他rhGH浓度也有促进作用,但与100 ng/ml相比作用减弱。加入浓度为10 ng/ml和100 ng/ml的rhGH 24小时后,GHR位点数量显著高于对照组,而10000 ng/ml组GHR位点数量显著降低。
不同浓度的rhGH对Bel-7402肝癌细胞系的生长可能产生不同影响。一定浓度的rhGH可能刺激该细胞系生长。rhGH可调节该细胞系中GHR的表达。
