Abstract

OBJECTIVE

To determine the effect of dihydroartiminisin on the proliferation and phosphorylation of mitogen-activated protein kinase (MAPK) in SKOV3 and OVCAR3 ovarian cancer cell lines.

METHODS

Methyl thiazolyl tetrazolium assay was performed to evaluate the anti-proliferative effect of dihydroartiminisin in SKOV3 and OVCAR3 cells, and Western blot was used to determine its effect on phosphorylation level of MAPK, including extra-cell regulated kinase (ERK) 1/2 and p38 protein kinase, in the two cell lines.

RESULTS

Dihydroartiminisin inhibited the proliferation of ovarian cancer cells in vitro, with a mean of 50% inhibition concentration (IC(50)) at 72 h of (9.0 +/- 1.4) micromol/L for SKOV3 and (5.5 +/- 1.2) micromol/L for OVCAR3 respectively. Compared to cells without dihydroartiminisin treatment, phosphorylation level of ERK 1/2 in SKOV3 and OVCAR3 cells treated with dihydroartiminisin decreased by 64.2% and 75.3% respectively (P < 0.05), while phosphorylation of p38 protein kinase in SKOV3 and OVCAR3 only decreased by 8.5%and 6.4%respectively (P > 0.05).

CONCLUSION

Dihydroartiminisin can inhibit the proliferation of ovarian cancer cell in vitro, probably through down-regulation of the phosphorylation of ERK 1/2 in ovarian cancer cells.

目的

确定双氢青蒿素对SKOV3和OVCAR3卵巢癌细胞系中丝裂原活化蛋白激酶(MAPK)增殖和磷酸化的影响。

方法

采用甲基噻唑基四氮唑蓝法评估双氢青蒿素对SKOV3和OVCAR3细胞的抗增殖作用,并用蛋白质免疫印迹法测定其对这两种细胞系中MAPK磷酸化水平的影响,包括细胞外调节激酶(ERK)1/2和p38蛋白激酶。

结果

双氢青蒿素在体外抑制卵巢癌细胞的增殖,在72小时时,SKOV3细胞的平均半数抑制浓度(IC50)为(9.0±1.4)μmol/L,OVCAR3细胞为(5.5±1.2)μmol/L。与未用双氢青蒿素处理的细胞相比,用双氢青蒿素处理的SKOV3和OVCAR3细胞中ERK 1/2的磷酸化水平分别降低了64.2%和75.3%(P<0.05),而SKOV3和OVCAR3中p38蛋白激酶的磷酸化仅分别降低了8.5%和6.4%(P>0.05)。

结论

双氢青蒿素可在体外抑制卵巢癌细胞的增殖,可能是通过下调卵巢癌细胞中ERK 1/2的磷酸化来实现的。
