Abstract

The detection of apoptosis based on the interaction between annexin V and phosphatidylserine is presented in this paper. The in vitro experimental results demonstrate that annexin V in polyethylenimine film can maintain its high affinity with phosphatidylserine translocated from the inner to the outer plasma membrane of the apoptotic cells, which may consequently inhibit the redox reaction of Ru(NH(3))(5)Cl at the annexin V and polyethylenimine comodified pyrolytic graphite electrode. Therefore, if Ca(2+), the required ion for the function of annexin V, is added into the test solution, the redox wave of Ru(NH(3))(5)Cl can be clearly changed to indicate apoptosis and a simple and convenient method to detect apoptosis is developed.

本文提出了一种基于膜联蛋白 V 与磷脂酰丝氨酸相互作用的细胞凋亡检测方法。体外实验结果表明,多聚赖氨酸修饰的金电极表面固定的膜联蛋白 V 仍能保持与从细胞内转移到细胞膜外的磷脂酰丝氨酸的高亲和性,从而抑制[Ru(NH3)5Cl]2+在膜联蛋白 V 和多聚赖氨酸共修饰的热解石墨电极上的氧化还原反应。因此,如果向测试溶液中加入细胞凋亡所必需的离子 Ca2+,[Ru(NH3)5Cl]2+/+的氧化还原峰将会明显改变,从而指示细胞发生凋亡。由此建立了一种简单、方便的检测细胞凋亡的方法。
