Abstract

BACKGROUND

The HLA-A02-B46 haplotype is one of most frequent haplotypes among Guangdong Han populations. To explore the characteristics of the HLA-A02-B46 haplotype in Guangdong Han populations, the genetic polymorphism of HLA-A02-B46 haplotype was analysed by PCR-SBT in our study.

FINDINGS

Among 88 samples with the homozygotes for HLA-A02-B46 in the low resolution, 4 different alleles for A02 (A0201, A0203, A0206, A0207) and 1 allele for B46 (B4601) were identified by PCR-SBT. Among them, the A0207 allele was the predominant allele. Inversely, among the samples with HLA-A2-B46(-), six alleles were detected for A02 (A020101, A0203, A0205, A0206 and A0207), and the A0201 allele was predominant. On the other hand, the HLA-A02-B46 haplotype presented moderate heterozygosis (32.95%). In addition, the linkage with DRB1 was analysed in HLA-A2-B46 haplotype, and there existed 10 alleles with DRB1. With the low resolution for DRB1, the other 10 DRB1 alleles all linked with the HLA-A02-B46 haplotype except for DRB101, DRB110, and DRB117. Moreover, we found eight alleles of DRB1 in the HLA-A0207-B4601 haplotype.

CONCLUSION

The polymorphism distribution of the A02 allele between the HLA-A02-B46 and HLA-A02-B46(-) haplotypes among the Guangdong Han populations provides useful information for research on unrelated hematopoietic stem cell transplantation (UHSCT), anthropology, and disease association for populations with the HLA-A02-B*46 haplotype.

HLA - A02 - B46单倍型是广东汉族人群中最常见的单倍型之一。为探索广东汉族人群中HLA - A02 - B46单倍型的特征,本研究采用聚合酶链反应 - 序列特异性引物分型法(PCR - SBT)分析了HLA - A02 - B46单倍型的基因多态性。

研究结果

在88例低分辨率下HLA - A02 - B46纯合子样本中,通过PCR - SBT鉴定出A02的4种不同等位基因(A0201、A0203、A0206、A0207)和B46的1种等位基因(B4601)。其中,A0207等位基因为主要等位基因。相反,在HLA - A2 - B46( - )样本中,检测到A02的6种等位基因(A020101、A0203、A0205、A0206和A0207),且A0201等位基因为主要等位基因。另一方面,HLA - A02 - B46单倍型呈现中度杂合性(32.95%)。此外,在HLA - A2 - B46单倍型中分析了与DRB1的连锁关系,存在10种DRB1等位基因。在DRB1低分辨率下,除DRB101、DRB110和DRB117外,其他10种DRB1等位基因均与HLA - A02 - B46单倍型连锁。此外,我们在HLA - A0207 - B4601单倍型中发现了8种DRB1等位基因。

结论

广东汉族人群中HLA - A02 - B46与HLA - A02 - B46( - )单倍型之间A02等位基因的多态性分布,为无关造血干细胞移植(UHSCT)研究、人类学以及HLA - A02 - B*46单倍型人群的疾病关联研究提供了有用信息。
