Abstract

A novel 3'-nucleotidase, which preferably hydrolyzed 3'-dTMP and 3'-dUMP, was isolated and purified from rabbit spleen cytosol. Since the main catalytic properties of the enzyme were similar to the rat liver nucleotidase reported by Fritzson and Smith, we also purified the latter enzyme from rat spleen. These two enzymes were different in several points: the substrate restriction, the molecular mass and the apparent charge. Further analyses with the antiserum raised in rat against the 3'-deoxyribonucleotidase evidenced that these enzymes were immunochemically different as well.

从兔脾脏细胞溶质中分离并纯化出一种新型的3'-核苷酸酶,该酶优先水解3'-dTMP和3'-dUMP。由于该酶的主要催化特性与Fritzson和Smith报道的大鼠肝脏核苷酸酶相似,我们也从大鼠脾脏中纯化了后一种酶。这两种酶在几个方面存在差异:底物特异性、分子量和表观电荷。用大鼠针对3'-脱氧核糖核苷酸酶产生的抗血清进行的进一步分析表明,这些酶在免疫化学上也不同。
