To investigate the effect of target therapy with cisplatin (CDDP)-loaded magnetic nanoparticles (MNP) in combination with chemoradiotherapy against nasopharyngeal carcinoma cell growth in nude mice.
Thirty-six BALB/c mice with implanted tumor of CNE-2 cells were randomly divided into 6 groups (n=6), including the control group, radiotherapy group, CDDP group, CDDP plus radiotherapy group, CDDP-MNP group, and CDDP-MNP plus radiotherapy group. The mice were given 0.3 ml normal saline (control) or corresponding agents (3 mg/kg) via the tail vein, and 0.5 ml saline was administered intragastrically before the injections. Before and after the treatment, the body weight, tumor volume and weight, and the tumor inhibition rates were measured. The proliferating cell nuclear antigen (PCNA) index was calculated on the basis of immunohistochemical staining.
Except for the cisplatin group, all the treated groups showed significantly reduced tumor volume as compared with that in the control group (P<0.05) with lowered body weight. Compared with the cisplatin group, the combined treatment groups showed significantly higher tumor inhibition rate (P<0.05), but the effect showed no significant difference from that in the radiotherapy group (P>0.05).
Targeted therapy with CDDP-loaded MNP alone or in combination with radiotherapy can effectively inhibit the growth of nasopharyngeal carcinoma in nude mice without increasing the toxicity.
探讨顺铂(CDDP)负载磁性纳米颗粒(MNP)靶向治疗联合放化疗对裸鼠鼻咽癌细胞生长的影响。
将36只接种CNE - 2细胞肿瘤的BALB/c小鼠随机分为6组(n = 6),包括对照组、放疗组、顺铂组、顺铂加放疗组、CDDP - MNP组和顺铂 - MNP加放疗组。小鼠经尾静脉注射0.3 ml生理盐水(对照组)或相应药物(3 mg/kg),注射前经胃管给予0.5 ml生理盐水。治疗前后测量体重、肿瘤体积和重量以及肿瘤抑制率。基于免疫组化染色计算增殖细胞核抗原(PCNA)指数。
除顺铂组外,所有治疗组的肿瘤体积均较对照组显著减小(P < 0.05),体重降低。与顺铂组相比,联合治疗组的肿瘤抑制率显著更高(P < 0.05),但与放疗组相比差异无统计学意义(P > 0.05)。
单独使用负载CDDP的MNP或联合放疗进行靶向治疗可有效抑制裸鼠鼻咽癌的生长,且不增加毒性。
