To investigate the dynamic changes of a-AR, b1-AR and b2-AR expression in hepatic fibrosis.
Rat hepatic fibrosis model was established by bile duct ligation (BDL). HE and Masson staining were used to determine hepatic fibrosis levels. Immunohistochemistry was applied to detect alpha -smooth muscle actin (alpha -SMA), a marker of hepatic stellate cell (HSC) activation; Western blot and real-time RT-PCR were used to measure the dynamic changes of alpha -AR, beta(1)-AR, beta(2)-AR expression on protein and mRNA levels, respectively, during the development of hepatic fibrosis.
(1) HE and Masson trichrome staining showed that the liver fibrosis models were established successfully. (2) At 1, 2, 3, 4 wk after BDL, alpha -SMA positive area density of the model group (10.58% +/- 1.75%, 24.14% +/- 2.02%, 29.74% +/- 2.59%, 34.28% +/- 2.01%) was significantly higher than that of the sham operation group (4.12% +/- 1.51%), P less than 0.01. (3) The expression of alpha -AR, beta(1)-AR, beta(2)-AR protein and mRNA was increased with the development of the hepatic fibrosis (P less than 0.05). (4) alpha -SMA expression was positively associated with alpha -AR, beta(1)-AR, beta(2)-AR, r values were 0.564, 0.753 and 0.606, respectively.
The expression of alpha -SMA is increased dramatically during the fibrosis, and is positively associated with the expression of alpha -AR, beta(1)-AR and beta(2)-AR.
探讨α-肾上腺素能受体(α-AR)、β1-肾上腺素能受体(β1-AR)和β2-肾上腺素能受体(β2-AR)在肝纤维化中的动态变化。
采用胆管结扎(BDL)法建立大鼠肝纤维化模型。采用苏木精-伊红(HE)和Masson染色法测定肝纤维化程度。应用免疫组织化学法检测肝星状细胞(HSC)活化标志物α-平滑肌肌动蛋白(α-SMA);采用蛋白质印迹法和实时逆转录-聚合酶链反应(RT-PCR)分别检测肝纤维化发展过程中α-AR、β1-AR、β2-AR蛋白和mRNA水平的动态变化。
(1)HE和Masson三色染色显示成功建立了肝纤维化模型。(2)BDL术后1、2、3、4周,模型组α-SMA阳性面积密度(10.58%±1.75%、24.14%±2.02%、29.74%±2.59%、34.28%±2.01%)显著高于假手术组(4.12%±1.51%),P<0.01。(3)随着肝纤维化的发展,α-AR、β1-AR、β2-AR蛋白和mRNA表达增加(P<0.05)。(4)α-SMA表达与α-AR、β1-AR、β2-AR呈正相关,r值分别为0.564、0.753和0.606。
在纤维化过程中α-SMA表达显著增加,且与α-AR、β1-AR和β2-AR的表达呈正相关。
