Abstract

A high performance reverse phase liquid chromatographic procedure is developed for simultaneous estimation of metformin hydrochloride and pioglitazone hydrochloride in combined tablet dosage form. The mobile phase used was a combination of acetonitrile:water:acetic acid (60:40:0.3) and the pH was adjusted to 5.5 by adding triethylamine. The detection of the combined dosage form was carried out at 230 nm and a flow rate employed was 1 ml/min. Linearity was obtained in the concentration range of 0.015 to 0.120 mug/ml of pioglitazone hydrochloride and 0.5 to 4.0 mug/ml of metformin hydrochloride with a correlation coefficient of 0.9992 and 0.9975. The results of the analysis were validated statistically and recovery studies confirmed the accuracy and precision of the proposed method.

开发了一种高效反相液相色谱法,用于同时测定复方片剂剂型中盐酸二甲双胍和盐酸吡格列酮的含量。所用流动相为乙腈:水:乙酸(60:40:0.3)的混合物,并通过加入三乙胺将pH值调至5.5。复方剂型的检测在230nm波长下进行,流速为1ml/min。盐酸吡格列酮浓度在0.015至0.120μg/ml范围内以及盐酸二甲双胍浓度在0.5至4.0μg/ml范围内呈线性关系,相关系数分别为0.9992和0.9975。分析结果经统计学验证,回收率研究证实了所提方法的准确性和精密度。
