Abstract

OBJECTIVE

To study the effects of repeated injections of betamethasone on proteoglycan concentration in the articular cartilage of normal knees of Californian rabbits of both sexes.

METHODS

Eighty animals were randomly divided into eight groups of ten animals each. Three control groups (saline solution injected or not) and five study groups - therapeutical doses, repeated or not, of betamethasone injected into the right knee of each animal at weekly intervals. After eight days from the last injection, sections of articular cartilage from tibial plateaus collected from weight-bearing surfaces were stained with hematoxylin and eosin for light microscopy analysis and with safranin O for the proteoglycan content assay. The staining intensity of safranin O was quantified by histomorphometry using an Olympus BX 50 microscope and a microcomputer with the Image Pro-plus 4.5O software.

RESULTS

Animals receiving one, two and four betamethasone injections showed no differences when compared to normal controls. Animals receiving six and eight injections had a significant decrease in safranin O staining intensity (p < 0.05) as compared to the control groups and the other study groups.

CONCLUSION

A decrease in the concentration of articular cartilage proteoglycans dependent on repeated betamethasone injection was effectively demonstrated.

目的

研究反复注射倍他米松对加利福尼亚雌雄兔正常膝关节软骨中蛋白聚糖浓度的影响。

方法

80只动物随机分为8组,每组10只。3个对照组(注射或不注射生理盐水)和5个研究组——每周一次向每只动物右膝注射治疗剂量的倍他米松,注射次数有重复或无重复。末次注射8天后,从负重面采集胫骨平台的关节软骨切片,用苏木精和伊红染色进行光学显微镜分析,用番红O染色测定蛋白聚糖含量。使用奥林巴斯BX 50显微镜和配备Image Pro-plus 4.5O软件的微型计算机,通过组织形态计量学对番红O的染色强度进行量化。

结果
