The therapeutic effect of sildenafil citrate on cerebral vasospasm after experimental subarachnoid hemorrhage (SAH) was studied in a rat model.
We used four groups of seven rats were as follows: no SAH, no treatment; SAH only; SAH plus 2 days of peroral sildenafil citrate 5mg/kg treatment and SAH plus 2 days of peroral sildenafil citrate 15 mg/kg treatment. Three different parameters were evaluated including the diameter of the basilar artery, the level of lipid peroxidation and the degree of the apoptosis 48 hours following SAH.
The results showed that sildenafil citrate attenuated SAH-induced cerebral vasospasm in the treatment groups in terms of the diameter of the basilar artery and lipid peroxidation in the two treatment groups, but there was no difference in terms of the level of apoptosis.
This study indicates that further research on the therapeutic effect of sildenafil citrate can be combined with the use of any apoptosis-blocking agent for the treatment of cerebral vasospasm following experimental subarachnoid hemorrhage.
在大鼠模型中研究枸橼酸西地那非对实验性蛛网膜下腔出血(SAH)后脑血管痉挛的治疗效果。
我们使用四组,每组七只大鼠,如下所示:未发生SAH,未治疗;仅SAH;SAH加2天口服5mg/kg枸橼酸西地那非治疗以及SAH加2天口服15mg/kg枸橼酸西地那非治疗。评估了三个不同参数,包括SAH后48小时基底动脉直径、脂质过氧化水平和细胞凋亡程度。
结果显示,在两个治疗组中,就基底动脉直径和脂质过氧化而言,枸橼酸西地那非减轻了SAH诱导的脑血管痉挛,但在细胞凋亡水平方面没有差异。
本研究表明,关于枸橼酸西地那非治疗效果的进一步研究可与使用任何凋亡阻断剂联合用于治疗实验性蛛网膜下腔出血后的脑血管痉挛。
