To investigate the effect of dimethylsulfoxide on the expression of thymic stromal lymphopoietin (TSLP) in human bronchial epithelial cell (HBE).
16HBE cells were incubated in the presence of dimethylsulfoxide at different concentrations, and the cell proliferation changes were observed. The expressions of TSLP mRNA and protein in the cells were detected by real-time quantitative PCR and ELISA, respectively.
Dimethylsulfoxide induced significantly increased TSLP mRNA expression in HBE cells (P<0.01) in a concentration-dependent manner. The level of TSLP protein in the supernatant was also increased after dimethylsulfoxide treatment, but high concentration of dimethylsulfoxide resulted in e inhibited cell proliferation.
Dimethylsulfoxide may affect the immunomodulatory function of HBE cells.
探讨二甲基亚砜对人支气管上皮细胞(HBE)中胸腺基质淋巴细胞生成素(TSLP)表达的影响。
将16HBE细胞在不同浓度的二甲基亚砜存在下孵育,观察细胞增殖变化。分别通过实时定量PCR和ELISA检测细胞中TSLP mRNA和蛋白的表达。
二甲基亚砜以浓度依赖性方式显著诱导HBE细胞中TSLP mRNA表达增加(P<0.01)。二甲基亚砜处理后上清液中TSLP蛋白水平也升高,但高浓度二甲基亚砜导致细胞增殖受抑制。
二甲基亚砜可能影响HBE细胞的免疫调节功能。
