To observe the effect of electroacupuncture (EA) at "Ciliao" (BL 32) on detrusor hyperreflexia and c-fos expression in the sacral segment of spinal cord in rats with spinal cord injury (SCI).
Thirty-seven adult female Sprague-Dawley rats were randomized into normal control (n=5), SCI model (n=16) and EA (n=16) groups. EA (20 Hz, 3 mA) was applied to bilateral BL 32 for 2 hours, once daily for 14 days. Intravesical pressure was detected by using a pressure transducer and a bioelectric amplifier. The expression of c-fos gene was detected by immunohistochemistry.
In comparison with normal control group, the maximum intravesical pressure (MIVP) raised significantly in model group (P < 0.05), and the compliance of the bladder decreased remarkably (P < 0.05). While compared with model group, MIVP decreased significantly in SCI rats of EA group after EA intervention (P < 0.05), and the vesical compliance increased obviously (P < 0.05). In comparison with normal control group, the mean optical density (OD) value of c-fos immuno-reaction (IR) positive products increased significantly in the sacral cord after SCI in model group (P < 0.05), while compared with model group, the mean OD value of c-fos IR positive products in EA group declined evidently but still being higher than that of normal control group (P < 0.05), displaying a downregulation of c-fos expression after EA.
Electroacupuncture at "Ciliao" (BL 32) can inhibit the overactivity of bladder in SCI rats and reduce the c-fos expression in the sacral cord, suggesting that the declined C-fibers' activity after EA may be one of its mechanism underlying improving detrusor hyperreflexia in spinal cord injury.
观察电针“次髎”(BL 32)对脊髓损伤(SCI)大鼠逼尿肌反射亢进及脊髓骶段c-fos表达的影响。
将37只成年雌性Sprague-Dawley大鼠随机分为正常对照组(n = 5)、SCI模型组(n = 16)和电针组(n = 16)。采用20 Hz、3 mA的电针刺激双侧BL 32,持续2小时,每日1次,共14天。使用压力传感器和生物电放大器检测膀胱内压。采用免疫组织化学法检测c-fos基因的表达。
与正常对照组相比,模型组膀胱最大内压(MIVP)显著升高(P < 0.05),膀胱顺应性显著降低(P < 0.05)。与模型组相比,电针干预后电针组SCI大鼠的MIVP显著降低(P < 0.05),膀胱顺应性明显增加(P < 0.05)。与正常对照组相比,模型组SCI后脊髓骶段c-fos免疫反应(IR)阳性产物的平均光密度(OD)值显著增加(P < 0.05);与模型组相比,电针组c-fos IR阳性产物的平均OD值明显下降,但仍高于正常对照组(P < 0.05),表明电针后c-fos表达下调。
电针“次髎”(BL 32)可抑制SCI大鼠膀胱过度活动,降低脊髓骶段c-fos表达,提示电针后C纤维活性降低可能是其改善脊髓损伤后逼尿肌反射亢进的机制之一。
