Abstract

OBJECTIVE

To investigate the effect of chondroitinase ABC (ChABC) on the expression of growth associated protein 43 (GAP-43) and glial fibrillary acidic protein (GFAP) after spinal cord injury (SCI) in rats.

METHODS

A total of 150 adult female SD rats, weighing 250-300 g, were randomly divided into ChABC treatment group (group A), saline treatment group (group B), and sham operation group (group C) with 50 rats in each group. In groups A and B, the rats were made the SCI models and were treated by subarachnoid injection of ChABC and saline; in group C, the rats were not treated as a control. At 1, 3, 7, 14, and 21 days after operation, the Basso, Beattie, and Bresnahan (BBB) score system was used to evaluate the motion function, and immunofluorescent histochemical staining was used to observe the expressions of GAP-43 and GFAP.

RESULTS

At different time points, the BBB scores of groups A and B were significantly lower than those of group C (P < 0.05); there was no significant difference in BBB score between groups A and B after 1, 3, and 7 days of operation (P > 0.05), but the BBB score of group A was significantly higher than that of group B after 14 and 21 days of operation (P < 0.01). At different time points, the GAP-43 and GFAP positive neurons of groups A and B were significantly higher than those of group C (P < 0.05). After 14 and 21 days of operation, the GAP-43 positive neurons of group A were more than those of group B (P < 0.01). After 7, 14, and 21 days of operation, the GFAP positive neurons of group A were significantly less than those of group B (P < 0.01).

CONCLUSION

ChABC can degrade glial scar, improve the microenvironment of the injured region and enhance the expression of GAP-43, which promotes axonal growth and extension.

探讨软骨素酶ABC(ChABC)对大鼠脊髓损伤(SCI)后生长相关蛋白43(GAP - 43)和胶质纤维酸性蛋白(GFAP)表达的影响。

方法

选取150只体重250 - 300 g的成年雌性SD大鼠,随机分为ChABC治疗组(A组)、生理盐水治疗组(B组)和假手术组(C组),每组50只。A组和B组大鼠制备SCI模型后,分别经蛛网膜下腔注射ChABC和生理盐水进行治疗;C组大鼠不做处理作为对照。术后1、3、7、14和21天,采用Basso、Beattie和Bresnahan(BBB)评分系统评估运动功能,采用免疫荧光组织化学染色观察GAP - 43和GFAP的表达。

结果

在不同时间点,A组和B组的BBB评分均显著低于C组(P < 0.05);术后1、3和7天,A组和B组的BBB评分差异无统计学意义(P > 0.05),但术后14和21天,A组的BBB评分显著高于B组(P < 0.01)。在不同时间点,A组和B组的GAP - 43和GFAP阳性神经元均显著多于C组(P < 0.05)。术后14和21天,A组的GAP - 43阳性神经元多于B组(P < 0.01)。术后7、14和21天,A组的GFAP阳性神经元显著少于B组(P < 0.01)。

结论
