Abstract

OBJECTIVE

To study the effects and mechanisms of Huanglian Jiedu Decoction (HJD) and its disassembled recipes containing serums on the proliferation and differentiation of preadipocytes.

METHODS

HJD and its disassembled recipes containing serums were prepared. The 3T3-L1 preadipocytes were cultured. The proliferation of 3T3-L1 preadipocytes was detected by methyl thiazolyl tetrazolium (MTT) method. The accumulation of lipid droplets in the cytoplasm of differentiated preadipocytes was observed by oil red O staining and quantitatively analyzed by colorimetry. The mRNA expressions of peroxisome proliferation activated receptor y (PPAR gamma) and CAAT/enhancer binding protein (C/EBP alpha) were detected by reverse transcription polymerase chain reaction (RT-PCR).

RESULTS

Intervention with serum containing HJD, Phellodendron amurense Rupr plus Gardenia jasminoides Ellis, or Gardenia jasminoides Ellis showed significantly stimulative effects on the proliferation of preadipocytes, as compared with that in the blank control group (P<0.05, P<0.01). The preadipocytes treated with serum containing HJD, Phellodendron amurense Rupr plus Gardenia jasminoides Ellis, Coptis chinensis Franch or Gardenia jasminoides Ellis showed that the lipid droplets in the cytoplasm were significantly lessened, so did the mRNA expressions of PPAR gamma and C/EBP alpha when compared with the blank control group (P<0.05, P<0.01).

CONCLUSIONS

HJD promoted the proliferation of preadipocytes, decreased the accumulation of lipid droplets during the differentiation of adipocytes, and inhibited the differentiation of adipocytes, which might be associated with its effects on decreasing the mRNA expressions of PPAR gamma and C/EBP alpha. Phellodendron amurense Rupr and Gardenia jasminoides Ellis were the main components of HJD playing these roles.

研究黄连解毒汤(HJD)及其含药血清拆方对前脂肪细胞增殖与分化的影响及作用机制。

方法

制备HJD及其含药血清拆方。培养3T3-L1前脂肪细胞。采用甲基噻唑基四氮唑蓝(MTT)法检测3T3-L1前脂肪细胞的增殖情况。用油红O染色观察分化的前脂肪细胞胞质内脂滴的积聚情况,并通过比色法进行定量分析。采用逆转录聚合酶链反应(RT-PCR)检测过氧化物酶体增殖激活受体γ(PPARγ)和CCAAT/增强子结合蛋白(C/EBPα)的mRNA表达。

结果

与空白对照组相比,含HJD血清、黄柏加栀子血清或栀子血清干预对前脂肪细胞的增殖有显著促进作用(P<0.05,P<0.01)。含HJD血清、黄柏加栀子血清、黄连血清或栀子血清处理的前脂肪细胞,其胞质内脂滴明显减少,PPARγ和C/EBPα的mRNA表达也较空白对照组降低(P<0.05,P<0.01)。

结论

HJD可促进前脂肪细胞增殖,减少脂肪细胞分化过程中脂滴的积聚,抑制脂肪细胞分化,这可能与其降低PPARγ和C/EBPα的mRNA表达有关。黄柏和栀子是HJD发挥这些作用的主要成分。
