The aim of this study was to investigate the antihypertensive effect of a peptide fraction (PepC) obtained from a whey protein concentrate following hydrolysis by Cynara cardunculus, as well as of its fraction with MW below 3 kDa (PepCF). Both these concentrates encompassed peptides that exhibited potent in vitro inhibition of angiotensin-converting enzyme (ACE): two were released from α-lactalbumin--KGYGGVSLPEW and DKVGINYW, and one from β-lactoglobulin--DAQSAPLRVY.
Upon oral administration, by gastric intubation, of 400 mg/kg body weight (bw) of those peptide concentrates, or 5 mg/kg bw of the corresponding synthetic peptides, to 12 wk-old spontaneously hypertensive rats (SHR), the systolic and diastolic blood pressures were monitored by the tail-cuff method--before, and 2, 4, 6, 8 and 24 h afterwards. Water and zofenopril (5 mg/kg bw)--a known ACE-inhibitor, were used as negative and positive controls, respectively. Acute administration of PepC, PepCF, KGYGGVSLPEW, DKVGINYW and DAQSAPLRVY caused antihypertensive effects in SHR; the maximum effect occurred by 4 h and 6 h after administration of the peptide concentrates and the synthetic peptides, respectively. PepC and KGYGGVSLPEW also showed ACE-inhibitory activity in vivo: the pressor effect of angiotensin I was significantly lower, and the response to bradykinin increased when the rats were pre-treated with either product.
Our results strongly suggest that PepC will be effective as nutraceutical ingredient for the formulation of functional foods aimed at hypertension control.
本研究旨在探讨乳清蛋白浓缩物经朝鲜蓟水解后得到的肽段(PepC)及其分子量低于 3 kDa 的肽段(PepCF)的降压作用。这两种浓缩物都包含具有强力体外抑制血管紧张素转化酶(ACE)作用的肽段:其中两种肽段由α-乳白蛋白释放——KGYGGVSLPEW 和 DKVGINYW,一种肽段由β-乳球蛋白释放——DAQSAPLRVY。
通过灌胃给予 12 周龄自发性高血压大鼠(SHR)400 mg/kg 体重(bw)的这些肽浓缩物,或 5 mg/kg bw 的相应合成肽,通过尾套法监测收缩压和舒张压——在之前、之后 2、4、6、8 和 24 小时。水和佐芬普利(5 mg/kg bw)分别作为阴性和阳性对照。急性给予 PepC、PepCF、KGYGGVSLPEW、DKVGINYW 和 DAQSAPLRVY 可引起 SHR 的降压作用;肽浓缩物和合成肽分别在给药后 4 小时和 6 小时达到最大效果。PepC 和 KGYGGVSLPEW 也在体内显示出 ACE 抑制活性:预先给予任一产品后,血管紧张素 I 的升压作用明显降低,缓激肽的反应增加。
我们的结果强烈表明,PepC 将作为功能性食品配方的营养成分有效控制高血压。
