To investigate the growth effects of 4-phenyl butyric acid (PBA) on human gastric carcinoma cells and their mechanisms.
Moderately-differentiated human gastric carcinoma SGC-7901 and lowly-differentiated MGC-803 cells were treated with 5, 10, 20, 40, and 60 μmol/L PBA for 1-4 d. Cell proliferation was detected using the MTT colorimetric assay. Cell cycle distributions were examined using flow cytometry.
The proliferation of gastric carcinoma cells was inhibited by PBA in a dose- and time-dependent fashion. Flow cytometry showed that SGC-7901 cells treated with low concentrations of PBA were arrested at the G₀/G₁ phase, whereas cells treated with high concentrations of PBA were arrested at the G₂/M phase. Although MGC-803 cells treated with low concentrations of PBA were also arrested at the G₀/ G₁ phase, cells treated with high concentrations of PBA were arrested at the S phase.
The growth inhibitory effect of PBA on gastric cancer cells is associated with alteration of the cell cycle. For moderately-differentiated gastric cancer cells, the cell cycle was arrested at the G₀ /G₁ and G₂/M phases. For lowly-differentiated gastric cancer cells, the cell cycle was arrested at the G₀/G₁ and S phases.
探讨 4- 苯基丁酸(PBA)对人胃癌细胞生长的影响及其作用机制。
用 5、10、20、40、60μmol/L 的 PBA 分别作用于人胃癌 SGC-7901 细胞和 MGC-803 细胞 1~4 d,MTT 比色法检测细胞生长情况,流式细胞术检测细胞周期分布。
PBA 对胃癌细胞的生长具有抑制作用,呈时间和剂量依赖性。流式细胞术显示,低浓度 PBA 作用的 SGC-7901 细胞被阻滞于 G₀/G₁ 期,高浓度 PBA 作用的 SGC-7901 细胞被阻滞于 G₂/M 期;低浓度 PBA 作用的 MGC-803 细胞也被阻滞于 G₀/G₁ 期,高浓度 PBA 作用的 MGC-803 细胞被阻滞于 S 期。
PBA 抑制胃癌细胞生长与细胞周期改变有关,对中分化胃癌细胞主要阻滞于 G₀/G₁ 和 G₂/M 期,对低分化胃癌细胞主要阻滞于 G₀/G₁ 和 S 期。
