To build up a method of efficiently transforming Isatis indigotica with the Agrobacterium.
Two kinds of Agrobocterium: ATCC 15834 and RI1601 were used to treat different fraction of Isatis indigotica. Explored the effect of cocultured, different antibiotic concentration and the media on multiply the hairy roots.
The explants with out coculture had had higher survival rate rooting rate and earlier sprout time. After ultrasonic treatment of plant, the indacement rate was two times than that of untreated one; The best antibiotic concentration was 400 mg/L; The proportion of the grow speed of the hairy root in the liquid culture media was 2 - 3 times than that of solid culture media, and 37 times of common roots.
The method is useful for establishing an efficiently transformatiem system of Isatis indigotica by Agrobacterium.
建立一种利用农杆菌高效转化菘蓝的方法。
使用两种农杆菌ATCC 15834和RI1601处理菘蓝的不同部位。探究共培养、不同抗生素浓度及培养基对毛状根增殖的影响。
未进行共培养的外植体具有较高的成活率、生根率和较早的发芽时间。对植株进行超声处理后,诱导率是未处理植株的两倍;最佳抗生素浓度为400mg/L;毛状根在液体培养基中的生长速度是固体培养基中的2至3倍,是普通根的37倍。
该方法有助于建立高效的农杆菌介导的菘蓝转化体系。
