Abstract

AIMS

Placental calcification is a common pathologic condition in obstetrics. To detect the bacteria infection mechanisms for calcification, an experiment was performed to isolate, culture, and identify the nanobacteria in placental calcification.

METHOD

Sixteen cases of placental calcification of pregnant women were collected for the purpose of the isolation of nanobacteria, cultivation, and identification of 16S rDNA sequence.

RESULT

Under transmission electron microscope, novel oval-shape nanobacteria-like particles (NLP) in extracellular matrix of calcified placenta tissues were found with 50-500 nm in diameter, and among hydroxyapatite crystals aggregation existed. After about 4 weeks of culturing and isolating NLP from these calcified tissues, all calcified placental tissue samples and one adjacent tissue of calcified placental tissue samples showed white granular depositions, which were firmly attached to the bottom of the culture tubes and visible to the naked eyes. In the control group they could not be seen. After PCR was amplified a 1407-bp fragment was obtained and submitted to GenBank after sequencing with accession number JN029830. The 16S rDNA sequence homology between the isolation strain and strain nanobacteria (X98418) was 92% in GenBank.

CONCLUSION

For the first time isolated, cultured, and identified nanobacteria in placental calcification indicated that nanobacteria infection is related to placental calcification.

目的

胎盘钙化是产科常见的病理状况。为探究钙化的细菌感染机制,开展了一项实验以分离、培养和鉴定胎盘钙化中的纳米细菌。

方法

收集16例孕妇胎盘钙化样本,用于纳米细菌的分离、培养及16S rDNA序列鉴定。

结果

在透射电子显微镜下,于钙化胎盘组织的细胞外基质中发现了新型椭圆形纳米细菌样颗粒(NLP),直径为50 - 500纳米,且存在于羟基磷灰石晶体聚集体中。从这些钙化组织中培养和分离NLP约4周后,所有钙化胎盘组织样本及1例钙化胎盘组织样本的相邻组织均出现白色颗粒状沉积物,它们牢固附着于培养管底部,肉眼可见。对照组则未见此类沉积物。经PCR扩增获得一个1407碱基对的片段,测序后提交至GenBank,登录号为JN029830。分离菌株与GenBank中纳米细菌菌株(X98418)的16S rDNA序列同源性为92%。

结论

首次在胎盘钙化中分离、培养并鉴定出纳米细菌,表明纳米细菌感染与胎盘钙化有关。
