To explore the effects of ezrin silencing on pancreatic cancer cell line Panc-1.
Pancreatic cancer cell line Panc-1 was transfected with ezrin silencing plasmid. The proliferation and the cell cycle status were determined by CCK-8 assay and flow cytometry analysis, respectively. Cellular membrane protrusions/microvilli formation were visualized by scanning election microscopy. Colony formation assay was used to determine the cell anchor-independent growth ability in vitro. Trans-filter migration and invasion assays were performed with 8 µm pore inserts in a 24-well BioCoat chamber with/without Matrigel.
Ezrin silencing decreased cellular protrusions/microvilli formation, anchorage-independent growth, cell migration and invasion, but had no effects on cell proliferation in vitro and cell cycle, in pancreatic cancer cell line Panc-1.

探讨埃兹蛋白沉默对胰腺癌细胞系Panc-1的影响。
用埃兹蛋白沉默质粒转染胰腺癌细胞系Panc-1。分别通过CCK-8法和流式细胞术分析测定细胞增殖和细胞周期状态。通过扫描电子显微镜观察细胞膜突起/微绒毛的形成。采用集落形成试验测定细胞体外非锚定依赖性生长能力。使用8μm孔径的Transwell小室在含/不含基质胶的24孔BioCoat培养板中进行跨膜迁移和侵袭试验。
在胰腺癌细胞系Panc-1中,埃兹蛋白沉默减少了细胞突起/微绒毛的形成、非锚定依赖性生长、细胞迁移和侵袭,但对体外细胞增殖和细胞周期无影响。
