This study aimed to investigate the genetic diversity of 174 isolates of symbiotic bacteria associated with Acacia melanoxylon obtained from 15 sampling sites in Guangdong, Fujian and Jiangxi provinces of China.
The 16S rDNA restriction fragment length polymorphism (RFLP) and phylogenetic analyses of the 16S rDNA and housekeeping genes (recA, glnII and atpD).
In the 16S rDNA PCR-RFLP analysis, 9 rDNA types were identified among the 174 isolates; Phylogenetic analyses based on 16S rDNA and housekeeping gene sequences indicated that 34 representative isolates belonged to the genus Bradyrhizobium, Rhizobium, Mesorhizobium in Alpha-Proteobacteria, and the most closely related strains are Bradyrhizobium liaoningense, Bradyrhizobium betae, Bradyrhizobium cytisi, Rhizobium multihospitium and Mesorhizobium plurifarium.
All of the isolates could be identified to general, and Bradyrhizobium, Rhizobium or Mesorhizobium could be the dominant microsymbiont. The microsymbionts associated with Acacia melanoxylon showed relative genetic diversity.
本研究旨在调查从中国广东、福建和江西三省15个采样点采集的174株与黑木相思共生的细菌的遗传多样性。
采用16S rDNA限制性片段长度多态性(RFLP)分析以及16S rDNA和看家基因(recA、glnII和atpD)的系统发育分析。
在16S rDNA PCR-RFLP分析中,174株分离物中鉴定出9种rDNA类型;基于16S rDNA和看家基因序列的系统发育分析表明,34株代表性分离物属于变形菌门α-变形杆菌纲中的慢生根瘤菌属、根瘤菌属和中生根瘤菌属,与之亲缘关系最近的菌株是辽宁慢生根瘤菌、甜菜慢生根瘤菌、刺槐慢生根瘤菌、多宿主根瘤菌和多源中生根瘤菌。
所有分离物均可鉴定到属水平,慢生根瘤菌属、根瘤菌属或中生根瘤菌属可能是优势微共生体。与黑木相思相关的微共生体表现出相对的遗传多样性。
