Abstract

PURPOSE

To evaluate the effect of placental growth factor (PlGF) gene knockdown in a murine model of laser-induced choroidal neovascularization.

METHODS

Choroidal neovascularization was induced in the left eyes of 11 mice by infrared laser. Small interfering RNA (siRNA, 20 picomoles/10 μl) corresponding to PlGF mRNA was administered intravitreally by Hamilton syringe in all subjects. One month later, fluorescein angiography and histolologic examination were performed.

RESULTS

No leakage was apparent in the 11 eyes treated with siRNA cognate to PlGF. The results of histological evaluation were consistent with angiographic findings showing absence of choroidal neovascularization.

CONCLUSION

Knockdown of the PlGF gene can inhibit the growth of laser-induced choroidal neovascularization in mice.

目的

评估胎盘生长因子(PlGF)基因敲低在激光诱导的脉络膜新生血管小鼠模型中的作用。

方法

用红外激光诱导11只小鼠左眼脉络膜新生血管形成。所有实验对象均通过汉密尔顿注射器经玻璃体腔内注射与PlGF mRNA对应的小干扰RNA(siRNA,20皮摩尔/10微升)。1个月后,进行荧光素血管造影和组织学检查。

结果

用与PlGF同源的siRNA处理的11只眼中未出现渗漏。组织学评估结果与血管造影结果一致,显示无脉络膜新生血管形成。

结论

PlGF基因敲低可抑制小鼠激光诱导的脉络膜新生血管生长。
