To investigate the expression of the stem cell marker Nanog in lung cancer tissues and the correlations between Nanog expression and clinic-pathologic characteristics as well as prognosis of lung cancer.
163 patients with lung cancers enrolled in the study. The expression of Nanog in the cell lines and lung cancers were evaluated by RT-PCR, immunofluorescence and immunohistochemisty. Then, the correlations between Nanog expression status and clinic-pathologic characteristics and prognosis of lung cancer patients were analyzed.
It showed that Nanog are higher expressed in lung cancer tissues compared to their normal counterparts in both mRNA and protein levels, and Nanog expression was observed to be positively correlated with tumor differentiation and clinical stages of lung cancer patients (P = 0.001 and 0.001). Nanog were mainly localized at the cytoplasm in the brown color in the lung cancers. In addition, nuclear staining of Nanog was more observed in poorly differentiated lung cancers compared to others (P = 0.01). Furthermore, survival analyses showed that over-expression of Nanog protein predicted a worse prognosis for lung cancer patients (P = 0.001).
Nanog can be an important prognostic marker for lung cancer, which may present a new therapeutic target for lung cancer patients in the future.

选取 163 例肺癌患者作为研究对象,采用 RT-PCR、免疫荧光和免疫组织化学方法检测 Nanog 在细胞系和肺癌组织中的表达,分析 Nanog 表达与肺癌患者临床病理特征和预后的关系。
结果表明,肺癌组织中 Nanog 的 mRNA 和蛋白表达水平均高于相应的正常组织,且 Nanog 的表达与肿瘤分化程度和临床分期呈正相关(P=0.001 和 P=0.001)。在肺癌组织中,Nanog 主要定位于细胞质,呈棕色。此外,与其他分化程度的肺癌相比,低分化肺癌中 Nanog 的核染色更为明显(P=0.01)。生存分析显示,Nanog 蛋白的过度表达预示着肺癌患者预后不良(P=0.001)。
Nanog 可能是肺癌的一个重要预后标志物,为未来肺癌患者的治疗提供了新的靶点。
