Abstract

Paired helical filaments (PHF) are unusual neuronal fibers which accumulate progressively in the brain in Alzheimer's disease (AD). The insolubility of PHF in various kinds of solvents enabled us to obtain highly purified PHF, but prevented the application of conventional analytical methods to identify their components. Here we report that antibodies against purified PHF recognize tau protein, a brain-specific microtubule-associated protein, suggesting that a portion of PHF is tau protein.

双螺旋丝(PHF)是一种异常的神经元纤维,在阿尔茨海默病(AD)患者的大脑中会逐渐积累。PHF在各种溶剂中均不溶解,这使我们能够获得高度纯化的PHF,但却阻碍了使用传统分析方法来鉴定其成分。在此我们报告,针对纯化的PHF的抗体可识别tau蛋白,一种脑特异性微管相关蛋白,这表明部分PHF是tau蛋白。
