Lysimachia capillipes Hemsl (Primulaceae), a folk medicinal plant in China, showed significant anti-tumor activity in recent studies. A reliable LC-MS/MS method was developed and validated for the simultaneous determination of capilliposide B and capilliposide C, the major bioactive components in this plant, in rat plasma.
Rat plasma and whole blood samples were pretreated with dichlorvos, an esterase inhibitor, minimizing degradation of analytes in biological samples. The method validation was conducted over the curve range of 10.0 to 5000 ng/ml for both analytes. The intra- and inter-day precision and accuracy of the QC samples showed ≤6.1% RSD and 1.3-3.7% relative error.
The method was successfully applied to determine the concentrations of capilliposide B and capilliposide C in incurred rat plasma samples, after administration of Lysimachia capillipes Hemsl extract for a rat PK study.
金钱草(报春花科)是中国的一种民间药用植物,近期研究显示其具有显著的抗肿瘤活性。建立并验证了一种可靠的液相色谱-串联质谱法,用于同时测定该植物主要生物活性成分毛茎点草苷B和毛茎点草苷C在大鼠血浆中的含量。
大鼠血浆和全血样品用酯酶抑制剂敌敌畏进行预处理,可最大程度减少生物样品中分析物的降解。两种分析物在10.0至5000 ng/ml的曲线范围内进行方法验证。质量控制样品的日内和日间精密度及准确度显示相对标准偏差≤6.1%,相对误差为1.3 - 3.7%。
该方法成功应用于在大鼠药代动力学研究中给予金钱草提取物后,测定大鼠血浆中毛茎点草苷B和毛茎点草苷C的浓度。
