At the early stages of the secretory cycle, the activation of secretory cell nuclear chromatin is observed: the binding of acridine orange increases, while chromatin condensation in the peripheral zone of the nucleus decreases. The increase of the chromatin transcriptional activity provides for a significant increase in cytoplasmic RNA. Under the action of oxytocin reaction product granules are accumulated in conglomerates suggesting some changes in calcium distribution in the decondensed chromatin.
