Abstract

OBJECTIVE

To observe the effect of Tetramethyl pyrazine (TMP) on the cytokines and inflammatory mediators in the serum and the synovial fluid of collagen-induced arthritis (CIA)rats, and further to investigate its possible mechanisms for treating rheumatoid arthritis (RA).

METHODS

Type II CIA rat model was established. Rats in the TMP group were administered with TMP at 50 mg/kg and 100 mg/kg, once daily. Dexamethasone at 2.0 mg/kg was intramuscularly injected to those in the Dexamethasone treated group, once daily. Normal saline at 2 mL/kg was given to those in the normal control group and the model group, once daily. All medication was started from the 7th day, lasting to the 35th day. CIA rats' foot swelling degree was observed. Contents of serum IL-1, IL-6, IL-2, NO and PGE2in the synovial fluid were detected by radioimmunoassay and nitrate reduction method.

RESULTS

Compared with the normal group, the foot swelling obviously increased, contents of NO and PGE2 in the synovial fluid were obviously elevated in the model group (P < 0.01). Compared with the model group, the foot swelling could be obviously inhibited by 100 mg/kg TMP and Dexamethasone; serum levels of IL-1 and IL-6 obviously decreased, serum IL-2 level obviously increased, contents of NO and PGE, decreased (P < 0.01). TMP 50 mg/kg could obviously inhibit the foot swelling of CIA rats (P < 0.01). There was no statistical difference in other indices (P > 0.05).

CONCLUSIONS

TMP at 100 mg/kg showed obvious inhibition on CIA rats. Its inhibitory effect might be correlated to inhibiting activities of endogenous cytokines and the generation of inflammatory mediators in inflammation local regions, improving contents of anti-inflammation cytokines, and inducing the balance of the inflammatory cytokine network.

目的

观察川芎嗪(TMP)对胶原诱导性关节炎(CIA)大鼠血清及关节滑液中细胞因子和炎症介质的影响,进一步探讨其治疗类风湿关节炎(RA)的可能机制。

方法

建立Ⅱ型CIA大鼠模型。TMP组大鼠分别按50mg/kg和100mg/kg给予TMP,每日1次。地塞米松治疗组大鼠肌内注射2.0mg/kg地塞米松,每日1次。正常对照组和模型组大鼠给予2mL/kg生理盐水,每日1次。所有药物均从第7天开始给药,持续至第35天。观察CIA大鼠足肿胀程度。采用放射免疫分析法和硝酸还原法检测血清白细胞介素-1(IL-1)、白细胞介素-6(IL-6)、白细胞介素-2(IL-2)、一氧化氮(NO)及关节滑液中前列腺素E2(PGE2)的含量。

结果

与正常组比较,模型组大鼠足肿胀明显增加,关节滑液中NO和PGE2含量明显升高(P<0.01)。与模型组比较,100mg/kg TMP和地塞米松可明显抑制大鼠足肿胀;血清IL-1和IL-6水平明显降低,血清IL-2水平明显升高,NO和PGE2含量降低(P<0.01)。50mg/kg TMP可明显抑制CIA大鼠足肿胀(P<0.01)。其他指标差异无统计学意义(P>0.05)。

结论

100mg/kg TMP对CIA大鼠有明显抑制作用。其抑制作用可能与抑制内源性细胞因子活性、减少炎症局部区域炎症介质生成、提高抗炎细胞因子含量、诱导炎症细胞因子网络平衡有关。
