Abstract

AIM

To investigate the association between LRP5 gene polymorphisms and response to alendronate in Chinese osteoporotic women.

MATERIALS & METHODS: Six hundred and thirty nine Chinese postmenopausal women with osteopenia or osteoporosis were included and received alendronate treatment. The A1330V polymorphism of LRP5 was investigated. Bone mineral density (BMD) and bone turnover markers (ALP and β-isomerized carboxy-telopeptide of type I collagen [β-CTX]) were measured before and after treatment. The correlation of LRP5 polymorphisms with changes in BMD and bone turnover biomarkers were analyzed after treatment.

RESULTS

After 12 months of treatment, participants with CC and CT genotypes had a larger increase in lumbar spine BMD and a larger decrease in serum β-CTX and ALP levels than those with TT genotype (all p < 0.001). No significant genotype-treatment interaction was found in hip BMD.

CONCLUSION

The A1330V polymorphism of LRP5 is possibly correlated with response to alendronate treatment in Chinese women with osteoporosis, and the TT genotype could possibly predict a weak response to alendronate.

目的

探讨 LRP5 基因多态性与中国绝经后骨质疏松妇女对阿仑膦酸钠反应的关系。

材料与方法

纳入 639 例患有骨质疏松症或骨量减少的中国绝经后妇女,给予阿仑膦酸钠治疗。检测 LRP5 的 A1330V 多态性。治疗前后测量骨密度(BMD)和骨转换标志物(碱性磷酸酶和Ⅰ型胶原羧基端肽β特殊序列[β-CTX])。分析治疗后 LRP5 多态性与 BMD 和骨转换标志物变化的相关性。

结果

治疗 12 个月后,CC 和 CT 基因型患者的腰椎 BMD 增加幅度更大,血清 β-CTX 和 ALP 水平降低幅度更大,与 TT 基因型相比(均 p<0.001)。髋部 BMD 无显著的基因型-治疗相互作用。

结论

LRP5 的 A1330V 多态性可能与中国骨质疏松症妇女对阿仑膦酸钠治疗的反应相关,TT 基因型可能预示着对阿仑膦酸钠的反应较弱。
