Abstract

OBJECTIVE

To investigate the association between expression of hepatocyte growth factor(HGF) and its receptor c-Met and primary colorectal cancers with synchronous liver metastases.

METHODS

A total of 30 colorectal cancer patients with synchronous liver metastasis underwent radical resection of primary cancer and liver cancer in our hospital from June 2001 to June 2010. According to lymphatic metastasis, patients were divided into group A(T1T4N1N2M1, n=21) and group B(T1T4N0M1, n=9). Twenty-one matched T1T4N1N2M0 and 21 T1T4N0M0 patients were used as the controls of group A. Nine matched T1~T4N0M0 patients were used as the controls of group B. Expressions of HGF and c-Met in tissues of primary loci, liver loci and metastatic loci were detected by immunohistochemistry.

RESULTS

In primary loci of group A, the positive rate of HGF was significantly higher than that of T1T4N1N2M0 and T1T4N0M0 controls [71%(15/21) vs. 43%(9/21), 19%(4/21), all P<0.05]. The positive rate of c-MET[90%(19/21)] was significantly higher compared to T1T4N0M0 control[43%(9/21), P<0.05], while not significantly different compared to T1T4N1N2M0 control[86%(18/21)]. In primary loci of group B, positive rates of HGF and c-MET were not significantly different as compared to T1~T4N0M0 control[6/9 vs. 5/9, P>0.05; 8/9 vs. 6/9, P>0.05]. Concordance of HGF and c-MET expression in group A among primary loci, lymphatic metastatic loci and hepatic metastatic loci was 81%(17/21) and 76%(16/21).

CONCLUSION

HGF-c-Met may play a role in colorectal cancer patients with synchronous liver metastasis who have regional lymphatic metastasis, and may have few effect on colorectal cancer with synchronous liver metastasis without corresponding lymphatic metastasis.

目的

探讨肝细胞生长因子(HGF)及其受体c-Met的表达与原发性结直肠癌伴同时性肝转移之间的关系。

方法

选取2001年6月至2010年6月在我院接受原发性癌和肝癌根治性切除术的30例结直肠癌伴同时性肝转移患者。根据淋巴转移情况,将患者分为A组(T1T4N1N2M1,n=21)和B组(T1T4N0M1,n=9)。选取21例匹配的T1T4N1N2M0患者和21例T1T4N0M0患者作为A组的对照。选取9例匹配的T1~T4N0M0患者作为B组的对照。采用免疫组织化学法检测原发灶、肝转移灶和转移灶组织中HGF和c-Met的表达。

结果

在A组原发灶中,HGF阳性率显著高于T1T4N1N2M0和T1T4N0M0对照组[71%(15/21)对43%(9/21)、19%(4/21),P均<0.05]。c-MET阳性率[90%(19/21)]显著高于T1T4N0M0对照组[43%(9/21),P<0.05],而与T1T4N1N2M0对照组[86%(18/21)]相比差异无统计学意义。在B组原发灶中,HGF和c-MET阳性率与T1~T4N0M0对照组相比差异无统计学意义[6/9对5/9,P>0.05;8/9对6/9,P>0.05]。A组中HGF和c-MET在原发灶、淋巴转移灶和肝转移灶中的表达一致性分别为81%(17/21)和76%(16/21)。

结论

HGF-c-Met可能在伴有区域淋巴转移的结直肠癌伴同时性肝转移患者中发挥作用,而对无相应淋巴转移的结直肠癌伴同时性肝转移可能影响较小。
