This study was purposed to investigate the RHCE gene polymorphisms in Chinese Jiangsu Han blood donors with and without RHD gene among serologic RhD negative population. PCR with sequence-specific primers (PCR-SSP) was used to detect RHCE genotype in 337 serologic RhD negative, RHD gene positive donors. The RHD gene-specific polymorphisms were also determined by PCR-SSP in these donors. The results showed that among 337 serologic RhD negative, RHD gene positive donor 20 were RHCEC/C, 62 RHCEC/c, 24 RHCEc/c, 25 RHCEE/e, and 81 RHCEe/e; the allele frequencies for RHCEC and RHCEc were 0.4811 and 0.5189, respectively; and for RHCEE and RHCEe 0.1179 and 0.8821. Among 231 RHD gene negative donors, 3 were RHCEC/C, 34 RHCEC/c, 194 RHCEc/c, 15 RHCEE/e, 216 RHCEe/e; the allele frequencies for RHCEC and RHCEc were 0.0866 and 0.9134, respectively; and allele frequencies for RHCEE and RHCEe were 0.0325 and 0.9675, respectively. It is concluded that the most prevalent allele of RHCE gene was RHCE*ccee in Chinese Han Jiangsu RHD gene negative population. There is statistical difference in RHCE genotype distribution among serologic RhD negative population with and without RHD gene.
本研究旨在调查中国江苏汉族血清学RhD阴性人群中有无RHD基因的献血者的RHCE基因多态性。采用序列特异性引物聚合酶链反应(PCR-SSP)检测337例血清学RhD阴性、RHD基因阳性献血者的RHCE基因型。同时用PCR-SSP测定这些献血者的RHD基因特异性多态性。结果显示,在337例血清学RhD阴性、RHD基因阳性献血者中,20例为RHCEC/C,62例为RHCEC/c,24例为RHCEc/c,25例为RHCEE/e,81例为RHCEe/e;RHCEC和RHCEc的等位基因频率分别为0.4811和0.5189;RHCEE和RHCEe的等位基因频率分别为0.1179和0.8821。在231例RHD基因阴性献血者中,3例为RHCEC/C,34例为RHCEC/c,194例为RHCEc/c,15例为RHCEE/e,216例为RHCEe/e;RHCEC和RHCEc的等位基因频率分别为0.0866和0.9134;RHCEE和RHCEe的等位基因频率分别为0.0325和0.9675。结论是,在中国江苏汉族RHD基因阴性人群中,RHCE基因最常见的等位基因为RHCE*ccee。血清学RhD阴性人群中有无RHD基因者的RHCE基因型分布存在统计学差异。
