This study aimed to explore the effects and mechanisms of polyunsaturated fatty acids in the endometrial carcinoma Ishikawa cells.
The Ishikawa cells were treated with n-6 PUFAs, n-3 PUFAs, or the mixture of n-6/n-3 PUFAs at a ratio of 1:1/10:1. Cell proliferation was quantified by MTT assay, while expressions of Akt, mTOR and VEGF mRNAs by PCR.
Treatment with n-6 PUFAs and 10:1 n-6/n-3PUFAs markedly promoted cell proliferation and up-regulated expressions of Akt, mTOR and VEGF mRNAs. Further, n-3 PUFAs and 1:1 n-6/n-3 PUFAs markedly suppressed cell proliferation and down-regulated expressions of Akt, mTOR and VEGF mRNAs.
Different ratios of n-6/n-3PUFAs exhibit differential effects on the endometrial carcinoma Ishikawa cells through the PI3k/Akt/mTOR signaling pathway and differentially regulate VEGF expression.
本研究旨在探讨多不饱和脂肪酸对子宫内膜癌 Ishikawa 细胞的作用及其机制。
用 n-6 多不饱和脂肪酸、n-3 多不饱和脂肪酸或 n-6/n-3 多不饱和脂肪酸以 1:1/10:1 的比例混合处理 Ishikawa 细胞。通过 MTT 法测定细胞增殖,通过 PCR 检测 Akt、mTOR 和 VEGF mRNA 的表达。
用 n-6 多不饱和脂肪酸和 10:1 的 n-6/n-3 多不饱和脂肪酸处理显著促进细胞增殖,并上调 Akt、mTOR 和 VEGF mRNA 的表达。此外,n-3 多不饱和脂肪酸和 1:1 的 n-6/n-3 多不饱和脂肪酸显著抑制细胞增殖,并下调 Akt、mTOR 和 VEGF mRNA 的表达。
