A novel bacterial strain THG-PC4 was isolated from soil sample of rice field and was characterized by using a polyphasic approach. Cells were Gram-negative, bright yellow colored, nonmotile and rod shaped. The strain was aerobic and catalase positive, optimum growth temperature, and pH were 28 °C and 7, respectively. On the basis of 16S rRNA gene sequence analysis, strain THG-PC4 belongs to the genus Lysobacter and is most closely related to Lysobacter fragariae KCTC 42236, followed by Lysobacter oryzae KCTC 22249, Lysobacter tyrosinelyticus KCTC 42235, Lysobacter terrae KACC 17646, Lysobacter yangpyeongensis KACC 11407, Lysobacter rhizosphaerae KCTC 42237 and Lysobacter niabensis KACC 11587. In DNA-DNA hybridization tests, the DNA relatedness between strain THG-PC4 and its closest phylogenetic neighbors was below 45 %. The DNA G + C content was 66.6 mol %, and the predominant respiratory quinone was ubiquinone-8. Flexirubin-type pigments were found to be present. The major cellular fatty acids were iso-C, iso-C ω9c, iso-C and iso-C 3OH. The predominant polar lipids were phosphatidylethanolamine and phosphatidylglycerol. The DNA-DNA hybridization, genotypic, chemotaxonomic and physiological data demonstrated that strain THG-PC4 represented a novel species within the genus Lysobacter, for which the name Lysobcater humi is proposed. The type strain is THG-PC4 (=KACC 18284 = CCTCC AB 2015292).
从稻田土壤样本中分离出一种新型细菌菌株THG-PC4,并采用多相分类法对其进行了鉴定。细胞呈革兰氏阴性,亮黄色,无运动性,杆状。该菌株为需氧菌,过氧化氢酶阳性,最适生长温度和pH分别为28℃和7。基于16S rRNA基因序列分析,菌株THG-PC4属于溶杆菌属,与草莓溶杆菌KCTC 42236关系最为密切,其次是水稻溶杆菌KCTC 22249、酪氨酸溶杆菌KCTC 42235、土壤溶杆菌KACC 17646、杨平溶杆菌KACC 11407、根际溶杆菌KCTC 42237和尼亚溶杆菌KACC 11587。在DNA-DNA杂交试验中,菌株THG-PC4与其最接近的系统发育邻居之间的DNA相关性低于45%。DNA G+C含量为66.6mol%,主要呼吸醌为泛醌-8。发现存在柔韧红素型色素。主要细胞脂肪酸为异-C、异-C ω9c、异-C和异-C 3OH。主要极性脂质为磷脂酰乙醇胺和磷脂酰甘油。DNA-DNA杂交、基因型、化学分类和生理数据表明,菌株THG-PC4代表溶杆菌属内的一个新物种,为此提出了Lysobcater humi这个名称。模式菌株为THG-PC4(=KACC 18284 = CCTCC AB 2015292)。
