Abstract

Rapid, accurate and sensitive ultra-performance liquid chromatography tandem mass spectrometry (UPLC-MS/MS) methods were developed and validated for the simultaneous quantitation of amoxicillin and clavulanic acid in human plasma and urine samples. Amoxicillin and clavulanic acid in both plasma and urine were extracted using a solid-phase extraction method. The compounds were separated on an Acquity UPLC HSS T3 column (2.1 × 100 mm, 1.8 μm). Ampicillin was used as the internal standard (IS) in plasma, while amoxicillin-d and sulbactam were used as ISs in urine. The lower limit of quantitation was 0.0500 and 0.0250 μg/mL for amoxicillin and clavulanic acid in plasma, and 0.0500 μg/mL for both analytes in urine. The established methods were validated in terms of selectivity, precision, accuracy, linearity, matrix effect, recovery, carryover, interaction, dilution integrity and stability, and successfully applied to a pharmacokinetic study of amoxicillin sodium and clavulanate potassium (10:1) injection in healthy volunteers.

建立并验证了快速、准确且灵敏的超高效液相色谱串联质谱(UPLC-MS/MS)方法,用于同时定量测定人血浆和尿液样本中的阿莫西林和克拉维酸。血浆和尿液中的阿莫西林和克拉维酸均采用固相萃取法进行提取。化合物在Acquity UPLC HSS T3柱(2.1×100 mm,1.8μm)上进行分离。血浆中使用氨苄西林作为内标(IS),而尿液中使用阿莫西林-d和舒巴坦作为内标。血浆中阿莫西林和克拉维酸的定量下限分别为0.0500和0.0250μg/mL,尿液中两种分析物的定量下限均为0.0500μg/mL。所建立的方法在选择性、精密度、准确度、线性、基质效应、回收率、残留、相互作用、稀释完整性和稳定性方面进行了验证,并成功应用于健康志愿者中阿莫西林钠和克拉维酸钾(10:1)注射液的药代动力学研究。
