Adoptive transfer of T lymphocytes is a useful technique to characterize the role of the immune system in hypertension and vascular disease. Here we describe as an example the isolation of splenic T regulatory cells from donor mice processed to obtain a single cell suspension, followed by negative and positive selection to obtain CD4 T cells and CD4CD25 Treg cells, respectively. Treg cells can be subsequently transferred to recipient animals.
