Reaction of {Ru(η-p-cymene)Cl} (1) with aminomethylphosphane derived from morpholine (P{CHN(CHCH)O} (A), PPh{CHN(CHCH)O} (B)) or piperazine (P{CHN(CHCH)NCHCH} (C), PPh{CHN(CHCH)NCHCH} (D)) results in four new piano stool ruthenium(II) coordination compounds: [Ru(η-p-cymene)Cl(A)] (2A), [Ru(η-p-cymene)Cl(B)] (2B), [Ru(η-p-cymene)Cl(C)] (2C) and [Ru(η-p-cymene)Cl(D)] (2D). Every complex was fully characterized using spectroscopic methods (H, C{H}, P{H} NMR and ESI-MS), elemental analysis, X-ray single crystal diffraction and DFT calculations. Preliminary studies of in vitro cytotoxicity on the A549 (human lung adenocarcinoma) and MCF7 (human breast adenocarcinoma) cell lines revealed 2A-2D activity in the same order of magnitude as in the case of cisplatin. Additionally, the study confirmed the ability of 2A-2D to interact with DNA helix and transferrin.
{Ru(η-对异丙基苯)Cl} (1) 与由吗啉衍生的氨甲基膦(P{CH₂N(CH₂CH₂)O} (A)、PPh{CH₂N(CH₂CH₂)O} (B))或哌嗪(P{CH₂N(CH₂CH₂)NCH₂CH₂} (C)、PPh{CH₂N(CH₂CH₂)NCH₂CH₂} (D))反应,生成四种新型的钢琴凳型钌(II)配位化合物:[Ru(η-对异丙基苯)Cl(A)] (2A)、[Ru(η-对异丙基苯)Cl(B)] (2B)、[Ru(η-对异丙基苯)Cl(C)] (2C) 和 [Ru(η-对异丙基苯)Cl(D)] (2D)。每种配合物均通过光谱方法(¹H、¹³C{¹H}、³¹P{¹H} NMR 和 ESI-MS)、元素分析、X 射线单晶衍射和 DFT 计算进行了全面表征。对 A549(人肺腺癌)和 MCF7(人乳腺腺癌)细胞系的体外细胞毒性初步研究表明,2A - 2D 的活性与顺铂的活性处于同一数量级。此外,该研究证实了 2A - 2D 与 DNA 螺旋和转铁蛋白相互作用的能力。
