Abstract

Icariin is a prenylated flavonol glycoside derived from the Chinese herb This study investigated the mechanism by which icariin prevents HO-induced apoptosis in rat nucleus pulposus (NP) cells. NP cells were isolated from the rat intervertebral disc and they were divided into five groups after 3 passages: (A) blank control; (B) 200 M HO; (C) 200 M HO + 20 M icariin; (D) 20 M icariin + 200 M HO + 25 M LY294002; (E) 200 M HO + 25 M LY294002. LY294002 is a selective inhibitor of the phosphoinositide 3-kinase (PI3K)/Akt signaling pathway. NP cell viability, apoptosis rate, intracellular reactive oxygen species levels, and the expression of AKT, p-AKT, p53, Bcl-2, Bax, caspase-3 were estimated. The results show that, compared with the control group, HO significantly increased NP cell apoptosis and the level of intracellular ROS. Icariin pretreatment significantly decreased HO-induced apoptosis and intracellular ROS and upregulated p-Akt and BCL-2 and downregulated caspase-3 and Bax. LY294002 abolished the protective effects of icariin. Our results show that icariin can attenuate H2O2-induced apoptosis in rat nucleus pulposus cells and PI3K/AKT pathway is at least partly included in this protection effect.

淫羊藿苷是一种来源于中国草药的异戊烯基黄酮醇苷。本研究探讨了淫羊藿苷预防过氧化氢(H₂O₂)诱导的大鼠髓核(NP)细胞凋亡的机制。从大鼠椎间盘分离出NP细胞,传代3次后将其分为五组:(A)空白对照;(B)200μM H₂O₂;(C)200μM H₂O₂ + 20μM淫羊藿苷;(D)20μM淫羊藿苷 + 200μM H₂O₂ + 25μM LY294002;(E)200μM H₂O₂ + 25μM LY294002。LY294002是磷酸肌醇3激酶(PI3K)/Akt信号通路的选择性抑制剂。评估NP细胞活力、凋亡率、细胞内活性氧水平以及AKT、p-AKT、p53、Bcl-2、Bax、caspase-3的表达。结果表明,与对照组相比,H₂O₂显著增加NP细胞凋亡和细胞内ROS水平。淫羊藿苷预处理显著降低H₂O₂诱导的凋亡和细胞内ROS,并上调p-Akt和BCL-2,下调caspase-3和Bax。LY294002消除了淫羊藿苷的保护作用。我们的结果表明,淫羊藿苷可减轻H₂O₂诱导的大鼠髓核细胞凋亡,PI3K/AKT通路至少部分参与了这种保护作用。
