Renal cell carcinoma (RCC) is one of the most common types of cancer in developed countries. Bone marrow stromal cell antigen 2 (BST2) gene, which encodes BST2 transmembrane glycoprotein, is overexpressed in several cancer types. In the present study, we analyzed the expression and function of BST2 in RCC.
BST2 expression was analyzed by immunohistochemistry in 123 RCC cases. RNA interference was used to inhibit BST2 expression in a RCC cell line.
Immunohistochemical analysis showed that 32% of the 123 RCC cases were positive for BST2. BST2 expression was positively associated with tumour stage. Furthermore, BST2 expression was an independent predictor of survival in patients with RCC. BST2 siRNA-transfected Caki-1 cells displayed significantly reduced cell growth and invasive activity relative to negative control siRNA-transfected cells.
These results suggest that BST2 plays an important role in the progression of RCC. Because BST2 is expressed on the cell membrane, BST2 is a good therapeutic target for RCC.
肾细胞癌(RCC)是发达国家最常见的癌症类型之一。编码BST2跨膜糖蛋白的骨髓基质细胞抗原2(BST2)基因在几种癌症类型中过表达。在本研究中,我们分析了BST2在肾细胞癌中的表达及功能。
采用免疫组织化学方法分析123例肾细胞癌病例中BST2的表达情况。运用RNA干扰技术抑制肾癌细胞系中BST2的表达。
免疫组织化学分析显示,123例肾细胞癌病例中有32%的BST2呈阳性。BST2表达与肿瘤分期呈正相关。此外,BST2表达是肾细胞癌患者生存的独立预测指标。与阴性对照siRNA转染的细胞相比,BST2 siRNA转染的Caki-1细胞的细胞生长和侵袭活性显著降低。
这些结果表明BST2在肾细胞癌进展中起重要作用。由于BST2在细胞膜上表达,因此BST2是肾细胞癌的一个良好治疗靶点。
