Raphanus sativus seeds are used as condiment and to treat hypogonadism, various ailments of liver and kidneys. The aim of this study was to evaluate the potential protective effects of methanol extract of R. sativus seeds (RSME) against hypogonadism induced with carbon tetrachloride (CCl) in Sprague-Dawley male rats. Thirty six rats were divided in to six groups with six animals in each. Animals of Group I were control and treated with saline, Group II, III and IV were given orally CCl (1 ml/kg bw; 10% in corn oil). Rats of Group III and IV were also simultaneously given RSME at 100 mg/kg bw and 200 mg/kg bw respectively. However, Group V and VI received RSME (100; 200 mg/kg bw, respectively) alone. All treatments were given at alternate days for 15 days. Treatment of CCl4 to rats decreased (P < 0.001) the level of CAT, POD, SOD, GST, GSH-Px and GSR antioxidant enzymes in testes of rat. Concentration of lipid peroxides (TBARS) was increased (P < 0.001) whereas concentration of GSH was decreased (P < 0.001) in testes of CCl4 treated animals. Concentration of testosterone, FSH and LH in serum was decreased (P < 0.001) while the level of estradiol and prolactin was increased (P < 0.001) in CCl4 treated rats. Injuries in seminiferous tubules were determined in histopathology of testes. Administration of RSME, dose dependently, markedly ameliorated the oxidative stress of CCl4 thereby restoring the level of antioxidant enzymes, lipid peroxides, reduced glutathione, male hormones and alterations in histopathology.
萝卜种子用作调味品,并用于治疗性腺功能减退以及肝脏和肾脏的各种疾病。本研究的目的是评估萝卜种子甲醇提取物(RSME)对四氯化碳(CCl)诱导的Sprague-Dawley雄性大鼠性腺功能减退的潜在保护作用。将36只大鼠分为六组,每组6只动物。第一组动物为对照组,用生理盐水处理;第二组、第三组和第四组口服CCl(1 ml/kg体重;10%溶于玉米油)。第三组和第四组的大鼠同时分别给予100 mg/kg体重和200 mg/kg体重的RSME。然而,第五组和第六组分别单独接受RSME(分别为100;200 mg/kg体重)。所有处理均隔天进行,共15天。给大鼠注射CCl4后,大鼠睾丸中CAT、POD、SOD、GST、GSH-Px和GSR抗氧化酶水平降低(P<0.001)。在注射CCl4的动物睾丸中,脂质过氧化物(TBARS)浓度升高(P<0.001),而GSH浓度降低(P<0.001)。在注射CCl4的大鼠中,血清中睾酮、FSH和LH浓度降低(P<0.001),而雌二醇和催乳素水平升高(P<0.001)。在睾丸组织病理学检查中确定了生精小管的损伤。RSME的给药剂量依赖性地显著改善了CCl4的氧化应激,从而恢复了抗氧化酶水平、脂质过氧化物、还原型谷胱甘肽、雄性激素水平以及组织病理学改变。
